Laser Capture Microscopy
The Advanced Microscopy & Imaging core facility houses a PIXCELL IIe
laser capture microscope (Arcturus Engineering) that can be used to
microdissect small clusters of cells, or even single cells, for use in
DNA, RNA or protein analyses. Cells can be dissected from embedded
tissue sections that have been stained with H&E, a special stain, or
even an immunostain.
Laser Capture Microscopy Procedure: The procedure is simple and rapid.
Training time is less than one hour, and a trained user can dissect
from one to several thousand cells within a matter of minutes.
The PIXCELL II scope has an integrated digital image capture unit,
allowing one to record "before" and "after" images of the dissected
cells. In the example shown below, a pure population of tumor cells
were dissected away from whole biopsy.
The Arcturus PixCell II Laser Capture Microdissection (LCM)
microscope. This instrument is attached to an Olympus IX 50 and is
equipped with a Sony DXC-151A Hyper HAD CCD/RIS/RGB camer with Sony
camera adaptor CMA-D2 . This system is also used independently to
capture images from H&E or HRP stained sections on glass slides . With
this instrument one can quickly locate a single cell or large group of
cells, technically aim-and-shoot and remove specific cells for
subsequent molecular analysis. Regardless of whether one uses
paraffin-embedded or frozen tissue samples, stained or immunolabeled
slides, the LCM preserves the exact morphologies of both the captured
cells as well as the surrounding tissue. The system is designed to
easily monitor and document the entire process and stores images in
the archiving workstation .

The Arcturus PixCell IIe Laser Capture Microdissection system is an
intuitive system for fast point-and-shoot microdissection. Cell
capture with the PixCell IIe LCM system is direct and assured. Cells
of interest can be directly picked up from the slide surface.
The laser capture micro-dissection microscope allows one to retrieve
or "capture" a selected cell or group of cells from a stained or
unstained paraffin or frozen section. Bio-molecules such as DNA, RNA,
or protein can then be isolated from the captured cells for
purification and analysis. Cells can also be captured from blood
smears and from fine needle aspirates.
This unit is based on the Olympus IX50 Inverted Microscope. It powers
on and looks like it's in beautiful condition. I don't have any slides
or experience to test this fully, but it looks like it's in good
shape. Has a 6-place turret. Only what is pictured is included in this
Listing.
One frequently used application of LCM is to obtain RNA from which
cDNA is made by RT-PCR and then used for micro-array analysis to
determine which genes are being expressed in the selected cells.
Isolation of DNA from cells or from individual chromosomes from
metaphase spreads can be used for genetic screening. Protein
aggregates can be accumulated from intracellular vesicles for
evaluation by western blot or amino acid Similarly, lipid droplets can
be captured for subsequent analysis using a variety of biochemical
techniques.
Laser capture microdissection (LCM) can be used to separate individual
cell types from within complex tissues and organs. DNA or RNA can then
be extracted from these microsections for gene expression analysis.
LCM utilizes a low-power infrared laser to melt a special
thermoplastic film over the cells or tissues of interest. Specifically
developed caps that are coated with this thermoplastic film are placed
on the tissue section of interest. The PixCell IIe LCM instrument is
then used to direct the low-powered infrared laser through the cap to
melt the film onto the cells of interest. These cells adhere to the
cap when it is lifted from the tissue section – thus, capturing them
for further analysis.
